Washington Life Sciences Discovery Fund (LSDF) has awarded nearly $4.7 million to launch Washington Vaccine Alliance (WAVA) supporting research into herpes vaccine. During the start-up phase, WAVA core operations will function within the Vaccine and Infectious Disease Institute of Fred Hutchinson Cancer Research Center. WAVA is a consortium of seven not-for-profit research institutions, focused on providing the skills and apparatus necessary to develop, test and deploy new or improved vaccines for many of the world's most challenging diseases, including herpes. Its structure will allow member organizations more effectively to develop and share critical skills and facilities, and partner with commercial pharmaceutical and biotech companies in herpes vaccine development and deployment. "The successful development and deployment of even one vaccine will have a significant impact on the health of the citizens and will provide a very cost-effective intervention to reduce overall health care costs." said Larry Corey, M.D., principal investigator of the LSDF-funded project and co-director of the Vaccine and Infectious Disease Institute at the Hutchinson Center. The LSDF award provides critical funding to complete the initial phases of three new vaccine projects, including the herpes vaccine and the ability to secure funding for the rest of the development process. The herpes vaccine projects aim is to develop and produce a vaccine for preliminary clinical trials. WAVA will create several core service centers, critical to bringing vaccines to market: discovery, development, production and deployment. These services will be extremely cost effective by taking advantage of existing expertise and sharing resources across the larger portfolio of vaccine projects. Member organizations will contract with each other to speed the testing and development process and The Hutchinson Center will oversee informatics and clinical trials design and management. "Washington State has accumulated a critical mass of expertise in crucial areas necessary for the successful identification and development of vaccines. The creation of WAVA provides the catalyst to allow each member to create new programs that build upon the substantial existing successes," said King Holmes, M.D., Ph.D., chair of the University of Washingtons Department of Global Health. The portfolio of vaccine projects for WAVA covers major infectious diseases that await the development of effective and affordable vaccines. These include HIV, sexually transmitted infections such as genital herpes, hepatitis C, influenza, diarrheal diseases (especially of infants and children), pneumonia in the elderly and the pandemic infections of tuberculosis and malaria.

We know by now how those painful and unsightly herpes cold sores around the mouth are caused by the herpes simplex virus type one (HSV-1). Some of us are lucky enough to be able to keep them at bay for long periods of time - and then some trigger like a fever, too much sunlight or stress in our lives causes them to come back all over again to give us more grief. That is because even when the herpes virus isnt actively punishing us with those horrid cold sores, it is still there, lurking in the depths where it can lay dormant sometimes for years. Now professor Bryan Cullen, professor of molecular genetics and microbiology at Duke University, Durham NC has published findings which may go some way to finding that elusive cure for herpes. The principle is this: in order to kill the herpes virus, you must first activate it. During non-active phases, the herpes HSV1 virus is present in the nervous system but does not reproduce itself. The same principle applies to the genitally transmitted herpes HSV2 virus and other latent viruses including the chickenpox virus. "Inactive virus is completely untouchable by any treatment we have. Unless you activate the virus, you can't kill it," says Professor Cullen. When the virus is in its dormant phase it does not reproduce, and produces only one molecular product, called LAT RNA, which had always proved something of a mystery to scientists. Professor Cullens research has shown that LAT RNA is processed into smaller strands known as micro RNAs. These micro RNAs prevent the virus from reproducing itself until such time as a major stress in the patient causes the herpes virus to start its manufacturing process over again. Professor Cullen and his team are currently testing a new drug which, if successful, will kick start the herpes virus by binding the micro RNAs. This will cause the herpes virus to reproduce. The patient would then be treated with the drug acyclovir which effectively kills replicating HSV1 viruses. "In principle, you could activate and then kill all of the virus in a patient. This would completely cure a person, and you would never get another cold sore. said Professor Cullen.

Researchers at Chicago’s Feinberg School of Medicine have debunked the myth that the HIV virus infects women only via areas of broken or damaged skin, such as herpes sores.

Until recently it had been assumed that the virus sought out herpes damaged skin in order to reach the immune system cells deeper in the tissue. Some scientists had even thought the normal (i.e. uninfected with herpes) lining of the vaginal tract offered protection from HIV infection during intercourse, aggravating an unpleasant stigma for herpes sufferers.

But Thomas Hope, Professor of cell and molecular biology at Northwestern University now admits: “Normal skin is vulnerable.” Professor Hope and others developed a new method of observing HIV by introducing a strain of the virus carrying fluorescent, light-activated tracers into vaginal tissue samples in a laboratory environment.

Scientists who hitherto had little understanding of how HIV infected women during intercourse were able to observe under the microscope how the virus was able quickly to penetrate the genital skin to reach immune cells. Professor Hope said the study indicated that the virus targets areas of skin which have recently shed skin cells and cast doubt on the previous theory that the virus required damaged or broken skin to penetrate.

He added that this could explain why a clinical trial in Africa using a diaphragm to block the cervix produced no reduction in HIV transmission and also why studies of drugs designed to prevent lesions in genital herpes had proved ineffective.

Emphasizing how this study underlines the need for vaccine treatments to prevent HIV infection, Professor Hope added: “People need to remember that they are vulnerable. The sad part is if people just used a condom, we wouldn’t have this problem.”

In the United States, HIV continues to be predominantly passed via male homosexual activity. Women account for 26 percent of all new HIV cases in the USA, according to the US Center for Disease Control and Prevention.

None of these participants had shown any signs or symptoms of oral herpes. This study used the most up-to-date techniques for herpes diagnosis. Therefore its finding can be considered as full proof that almost all adult population has oral herpes whether they are aware of it or not. This is why it is really important to remember this while dealing with newborn babies.

The WHO reports that women are at greater risk of catching herpes than men.

A sadistic and resentful ex-boyfriend distributed pornographic fliers displaying his ex having sex with disease carrier written across because he knew she had herpes. He just couldn't handle being dumped or so it seemed so he started faxing these fliers at his ex girlfriend workplace and outside her son's middle school as revenge. He's now being prosecuted for giving that information out. I hope he goes to jail and gets to pay a huge fee. What saddens me most is that he didn't care for her ex poor son who has been traumatized for seeing his mother displayed like that. Who thinks about the kids? It seems not enough people. The researchers further say they pinpointed six specific genes in that chromosomal region as candidates for making people prone to outbreaks of cold sores (also called "fever blisters"). Cold sores occur when the herpes virus reactivates from its inactive state within the nerve, infecting the lip, nose, or face. This discovery is a breakthrough in the eternal fight against Herpes. The researches traced genetic markers in families to identify regions on chromosomes that harbor disease genes. Recent studies have suggested that Acyclovir, a general medicine used to treat the commonly found herpes simplex virus-2 (HSV-2), does not decrease the risk of HIV infection in people infected with HSV-2. The Phase III clinical trial was led by the University of Washington in Seattle, in coordination with the HIV Prevention Trials Network, an international consortium funded by the National Institute of Allergy and Infectious Diseases (NIAID) in the National Institutes of Health. The earlier researches had indicated that patients infected with HSV-2 are at a greater risk of contracting HIV. Scientists had hoped that due to its ability to suppress Herpes Virus and the associated genital sores and breaks in the skin, acyclovir could reduce the possibility of HIV transmission via sexual intercourse with a HSV-2 infected person. Although Acyclovir did not help in the risk reduction of HIV, Dr. Connie Celum, the committee chairperson, indicated that acyclovir did reduce the genital ulcers associated with HSV-2. He also indicated that now the committee will concentrate on researching the factors which make HSV-2 a risk factor for HIV. My comment: I'm not surprised. I read shortly after a reassuring article saying that it didn't mean that herpes medication didn't reduce herpes symptoms or shedding. However I don't see any other way. This study could mean that genital herpes medicines fail to prevent micro-ulcerations caused by very mild herpes symptoms and in other words fail to protect from other STDs. This could also contradict the previous findings saying that daily herpes medical treatment did reduce the probability of herpes infection. If it does, it should logically also have an impact on other STD transmission. To me the only product that works today is a local ointment. I'll tell all about it in the second edition of Herpes Wise.
